The Chemistry of Human Epidermis

چکیده

Recently information has been presented concerning the close chemical relationship of the proteins of many ectodermal tissues. It has become increasingly clear that the keratins of these structures have a similar molecular pattern and may be tentatively defined as proteins which are resistant to digestion by pepsin and trypsin, which are insoluble in dilute acids, dilute alkalies, water, and inorganic solvents, and which on acid hydrolysis yield such quantities of histidine, lysine, and arginine that the approximate molecular ratios of these amino acids are respectively 1:4: 12 (Block and Vickery, 1931). It was significant that in these studies, which included among other keratins human hair and finger nails, no data concerning human epidermis were presented. Since histologically the nails and hair are considered as specialized and highly keratinized appendages of the epidermis, an analysis of this tissue might afford another chemical confirmation of the course of development. The material analyzed consisted of the layer of epidermis known as the stratum corneum. It was obtained from a patient having a subacute inflammatory condition of the skin diagnosed as dermatitis exfoliativa, a disease characterized by an abundant flaky desquamation of the upper two-thirds of the stratum corneum. Normally the cells of this area are dead, extremely desiccated, and partially keratinized but desquamation is so imperceptible as to make impossible the collection of a sample adequate for study.

Biochemical Methods in Production of Three-Dimensional Scaffolds from Human Skin: A Window in Aesthetic Surgery

BACKGROUND Use of matrix-derived biologic scaffolds has become a treatment of choice in several clinical issues. This study assessed biochemical methods in production of three-dimensional scaffolds from human skin. METHODS Human skin was prepared from circumcisions, washed in phosphate buffer saline (PBS) and kept at -20ºC until use. The skin samples underwent various methods. In group A, Na...

در ملانوم بدخیم Consumption of the epidermis

Background and Aim: Consumption of the epidermis (COE) is defined as thinning of the epidermis with attenuation of the basal and suprabasal layers and loss of rete ridges in areas of direct contact with malignant melanocytes. The aim of this study was to investigate the importance of COE as an additional diagnostic criterion for malignant melanoma and to evaluate its relationship to clinicopath...

Identification and cloning of a new calmodulin-like protein from human epidermis.

After separating by two-dimensional gel electrophoresis an extract of total proteins from human stratum corneum, two spots were extracted and analyzed for their peptide sequence. The resulting internal protein sequences provided evidence for the identification of a new calcium-binding protein. Cloning of the corresponding full-length cDNA was achieved by reverse transcriptase-polymerase chain r...

Establishment of a Primary Cell Culture of Human Fibroblast in Iran

Background: Human fibroblasts are the part of the dermis that secrete extracellular matrix for the purpose of tissue repair. Culturing fibroblasts, which leads to formation of a monolayer of these cells, is used for treating various conditions including thermal burns and other skin defects such as diabetic and varicose vein leg ulcers. Therefore, we aimed at developing a fibroblast bank to acco...

Evaluation of Wi-Fi Radiation Effects on Antibiotic Susceptibility, Metabolic Activity and Biofilm Formation by Escherichia Coli 0157H7, Staphylococcus Aureus and Staphylococcus Epidermis

Background: The radiation emitted from electromagnetic fields (EMF) can cause biological effects on prokaryotic and eukaryotic cells, including non-thermal effects. Objective: The present study evaluated the non-thermal effects of wireless fidelity (Wi-Fi) operating at 2.4 GHz part of non-ionizing EMF on different pathogenic bacterial strains (Escherichia coli 0157H7, Staphylococcus aureu...

Probing the Binding of Valacyclovir Hydrochloride to the Human Serum Albumin

UV-visible and Fluorescence spectroscopic methods were employed to study the interaction of human serum albumin (HSA) with Valacyclovir Hydrochloride. Additionally, molecular dynamics and molecular docking simulations were used to visualize and specify the binding site of Valacyclovir Hydrochloride. The Stern-Volmer and van't Hoff equations along with spectroscopic observations, were used to de...
